报告摘要:
G-protein coupled receptors (GPCRs), including opioid receptors, play critical roles in neuromodulation. It remains a major challenge to record or manipulate GPCR signaling events both with high spatial resolution and across a large brain volume. Fast Scan Cyclic Voltammetry and microdialysis significantly enabled the study of neuromodulator release events in the brain, but do not provide a cellular-resolution map of neuromodulators. GPCR-based genetically-encoded real-time sensors are transformative by enabling the investigation of neuromodulators with high spatiotemporal resolution. However, real time imaging is often times constrained to monitoring a small brain region, and does not allow further interrogation of the neuronal population in study. My research group focuses on designing integration sensors that can integrate the neuromodulation signal into a permanent mark in the cells for further investigation. We have designed an opioid sensor with the potential for whole-brain mapping. We are also extending this design to detect other neuromodulators across the brain. Additionally, we are designing genetically-encoded chemical-activated opioid peptides to enable cell type-selective manipulation of opioid receptors to study their causal-effect on pain modulation and addition. Lastly, we are actively involved in developing protein-based biologics as research tools and potential therapeutics for targeting protein aggregates in neurodegenerative diseases.

王文静,密歇根大学生命科学院与化学系助理教授,2006年毕业于澳门沙金在线平台,2012年在密歇根州立大学(导师Babak Borhan)获得博士学位。之后在2012-2018麻省理工和斯坦福大学开展博士后研究(导师: Alice Ting)。2018年起任职于密歇根大学,总体研究目标是利用蛋白工程和进化来设计一系列的分子探针和工具来研究神经调节物质和神经系统,和神经退行性疾病。目前已以第一或通讯作者在 Science, Nature Biotechnology, Nature Chemical Biology, Nature Methods, Nature Communications, Journal of the American Chemical Society, Angew Chem Int Ed, ACS Chembio等国际重要学术期刊上发表论文20余篇,申请国际发明专利8项。基于她实验室的工作,她近期获得了NIH Director’s New Innovator Award (2022) 和 Camille Dreyfus Teacher Scholar Award (2023).
